Scope of Work

The requirement involves DNA sequencing of PCR amplicons, ranging between 100-400 bp, derived from environmental DNA extracted from filtered seawater samples. The primary objective is to identify the taxonomic composition of these samples.

Key Requirements

  • Technology: Sequencing must be conducted on an Illumina NovaSeq 6000 using a 2x250 bp configuration on SP flowcells to maintain analytical consistency.
  • Quality: A minimum of 75% of bases must be ≥Q30.
  • Experience: The provider must have 15 or more years of experience in genomic shotgun and amplicon sequencing services.
  • Track Record: A demonstrated record of project completion through sequencing environmental DNA amplicons on Illumina NovaSeq 6000 SP flowcells, including for federal government research laboratories, is required.
  • Operational: The provider must perform 100% of the requested sequencing service in-house. This includes performing quality control (QC) on provided sequencing libraries and notifying the principal investigator within 72 hours of receiving damaged libraries or if input material fails QC. Demultiplexed fastq files must be provided after each run with sufficient time for data download.
